Gabapentin is commonly used to treat neuropathic pain (pain due to nerve damage). This review updates a review published in 2014, and previous reviews published in 2011, 2005 and 2000. To assess the analgesic efficacy and adverse effects of Gabapentin binds to the α2δ-1 subunit of voltage-gated calcium channels in the CNS, particularly in presynaptic neurons. The inhibition of neurotransmitter release leads to dampened neuronal hyperexcitability, especially in epileptic foci and pain pathways. Gabapentin blocks the tonic phase of nociception induced by formalin and carrageenan, and exerts a potent inhibitory effect in neuropathic pain models of mechanical hyperalgesia and mechanical/thermal allodynia. Neuropathic pain: The recommended gabapentin dosage ranges from 300 to 1200 mg taken orally 3 times daily, with a maximum daily dosage of 3600 mg. Fibromyalgia: The recommended gabapentin dosage is between 400 and 800 mg taken orally 3 times daily, with a maximum daily dosage of 2400 mg.
